<バキュロウイルスを用いた遺伝子導入ベクターの開発>
 本研究室では、ヒト、マウス等の高等哺乳動物細胞への遺伝子導入法としてバキュロウイルスを応用したベクターの開発を行っている。バキュロウイルスコートタンパク質の代わりに哺乳類細胞の細胞表面タンパク質に親和性の強いタンパク質を発現、提示し、効率よくウイルス粒子を細胞表面に吸着させ、遺伝子導入するベクターである。
現在のところ、異なる細胞種ごとに遺伝子導入可能な方法は確立していない。そこで細胞種特異的に発現するレセプターなどに対するリガンドやホーミングペプチドなどをバキュロウイルス本来のスパイクタンパク質と融合させたものをウイルス表面に発現、提示させることにより特定の細胞をターゲットにして選択的に細胞表面に吸着させ、遺伝子導入するベクター系 「ターゲットベクター」を開発することを目標としている。
また、ガンや白血病細胞で特異的に発現する細胞表面マーカーに対する抗体を提示したベクターなどを開発し、がん細胞に細胞死を引き起こす遺伝子を導入して病変細胞を特異的に除去するベクターの開発も試みている。

Development of the versatile expression vectors using Baculovirus

Genetics using gene transfer and ectopic gene expression is one of the most prevailing technologies to investigate the functions of gene products. Many attempts to apply various viruses to vehicles for the gene delivery have been performed. However, it has not been established yet to deliver genes in the cell type specif ic manner.

Our ongoing project is to develop the viral vectors which may target specif ic or limited cell types, such as blood cells or neurons, by modifying the genome of Autographa californica multiple nucleopolyhedrovirus (AcMNPV).

The baculovirus Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is an insect virus possessing a 134-kb double-stranded circular DNA genome packaged into a rod-shaped capsid.

The expression system using strong polyhedrin promoter has high ef f iciency in insect cells, and expression products receive eukaryotic post-translational modif ication, where biological activity may be held, thus baculoviral vector has been commonly used for the production of recombinant proteins.

Baculovirus is reported to show competency to infect mammalian cells such as hepatic cells, when the viral titer is extremely high. Since baculovirus is replication defective in mammalian cells, the possibility to utilize baculovirus as a safe vector for gene delivery in mammalian cells is seriously concerned.

Generally, viruses initiate infection to a cell by adsorption to specif ic membrane molecules, such as phospholipids, glycoproteins or receptors on the cell surface.

And it was reported that several mammalian viral envelope proteins (such as vesicular stomatitis virus G glycoprotein, VSV-G) expressed ectopically on the viral surface, remarkably increase the ef f iciency of viral infectivity and gene delivery to mammalian cells.

The purpose of our research work is to improve this vector system for ef f icient gene delivery and therapy targeting specif ic mammalian cell types.

We are now trying to construct baculoviral vectors which can display molecules such as ligand or antibodies which recognize mammalian membrane proteins specif ic for cell types.
